Variable masses in fission and heavy-ion collisions
With the use of the cranking formula, the coordinate-dependent mass parameters of the kinetic-energy operator in fission processes and heavy-ion collisions are calculated in the two-center oscillator model. It is shown that the reduced mass and also the classical moment of inertia are obtained for large separations of the fragments. For small separations, however, the mass parameter for the motion of the centers of mass of the fragments is larger than the reduced mass by an order of magnitude
Development of a fully coupled biogeochemical reactive transport model to simulate microbial oxidation of organic carbon and pyrite under nitrateâreducing conditions
©2018. American Geophysical UnionIn regions with intensive agriculture nitrate is one of the most relevant contaminants in groundwater. Denitrification reduces elevated nitrate concentrations in many aquifers, yet the denitrification potential is limited by the concentration of available electron donors. The aim of this work was to study the denitrification potential and its limitation in natural sediments. A column experiment was conducted using sediments with elevated concentrations of organic carbon (total organic carbon 3,247 mg C/kg) and pyrite (chromium reducible sulfur 150 mg/kg). Groundwater with high nitrate concentration (100 mg/L) was injected. Measurements were taken over 160 days at five different depths including Nâ and Sâisotope analysis for selected samples. A reactive transport model was developed, which couples nitrate reduction with the oxidation of organic carbon (heterotrophic denitrification) and pyrite (autolithotrophic denitrification), and considers also transport and growth of denitrifying microbes. The denitrification pathway showed a temporal sequence from initially heterotrophic to autolithotrophic. However, maximum rates were lower for heterotrophic (11 mmol N/(L*a)) than for autolithotrophic denitrification (48 mmol N/(L*a)). The modeling showed that denitrifying microbes initially preferred highly reactive organic carbon as the electron donor for denitrification but were also able to utilize pyrite. The results show that after 160 days nitrate increased again to 50 mg/L. At this time only 0.5% of the total organic carbon and 46% of the available pyrite was oxidized. This indicates that denitrification rates strongly decrease before the electron donors are depleted either by a low reactivity (total organic carbon) or a diminishing reactive surface possibly due to the presence of coatings (pyrite)
CO2 sequestration in deep aquifers
Journal ArticleDisposal and long-term sequestration of anthropogenic "greenhouse gases" such as CO2 is a proposed approach to reducing global warming. Deep, regional-scale aquifers in sedimentary basins are possible sites for sequestration, given their ubiquitous nature. We used a mathematical sedimentary basin model, including multiphase flow of CO2, groundwater, and brine, to evaluate residence times in possible aquifer storage sites and migration patterns and rates away from such sites in the Powder River Basin of Wyoming. We also used the model to simulate CO2 flow through fractures, to evaluate partitioning between fracture and rock matrix. These simulations provide insight regarding the ultimate propensity of permeability reductions versus permeability increases in the fracture zone associated with carbonate reactions. Regional-scale hydrologic properties, including the presence of fracture zones, were calibrated using surface heat flow data. Our initial results suggest that, in general, long-term (~1000 years or more) sequestration in deep aquifers is possible, if subsurface structure and permeability are well characterized. However, additional risks are involved. In addition to CO2 escape from sequestration aquifers into other aquifers or to the land surface, another environmental threat posed by subsurface sequestration is contamination by brines. We evaluated the potential for such unintended aquifer contamination by displacement of brines out of adjacent sealing layers such as marine shales. Results suggest that sustained injection of CO2 may incur wide-scale brine displacement out of adjacent sealing layers, depending on the injection history, initial brine composition, and hydrologic properties of both aquifers and seals
Regional-scale permeability by heat flow calibration in the Powder River Basin, Wyoming

Forward modeling of coupled fluid and heat flow in the Powder River basin, Wyoming, is used to explain anomalously high heat flow values observed in the southern portion of the basin. Effective basin-scale permeabilities of selected Powder River basin aquifers and aquitards were calibrated by matching surface heat flow measurements to simulation results. Fractures associated with a large anticline in the southwestern part of the basin were found to play a major role in the basin's thermal regime. While the model results are non-unique, they demonstrate that regional structural features play an important role in a basin's overall energy budget and fluid flow regime. Identifizierung und Charakterisierung von Kandidatengenen aus der DiGeorge Syndrom-Region auf dem kurzen Arm von Chromosom 10
Die vorliegende Promotionsarbeit beschĂ€ftigt sich vorwiegend mit der Untersuchung von Patienten mit partieller Monosomie 10p. Der PhĂ€notyp dieser Patienten Ă€hnelt hĂ€ufig dem des DiGeorge-Syndroms. Neben fazialen Dysmorphien und weiteren Nebensymptomen ist die Symptomentrias Herzfehler, T-Zelldefekt und Hypoparathyreoidismus das typische Merkmal dieses Entwicklungsdefektes. Viele Patienten mit einer partiellen Monosomie 10p zeigen diese Symptome, was fĂŒr einen DiGeorge-Syndrom-Locus auf Chromosom 10p spricht. Der Hauptlocus fĂŒr das DiGeorge-Syndrom liegt jedoch auf Chromosom 22q11. Mehr als 90 % der DiGeorge-Syndrom-Patienten haben eine Mikrodeletion 22q11. Diese Mikrodeletion zĂ€hlt mit einer Frequenz von etwa 1/4000 zu den hĂ€ufigsten Deletionen beim Menschen ĂŒberhaupt und ist deshalb schon seit langem das Ziel intensiver ForschungstĂ€tigkeit. Dennoch ist es erst in der jĂŒngsten Zeit gelungen, zumindest ein Gen aus der Mikrodeletionsregion 22q11 (TBX-1) zu isolieren, welches fĂŒr den beobachteten Herzfehler verantwortlich sein könnte. Ansonsten sind die molekularen Ursachen dieses Entwicklunsdefektes noch immer weitgehend unbekannt. Die Deletionen auf Chromosom 10p sind sehr selten. Sie sind aber von wissenschaftlichem Interesse, da die molekulare AufklĂ€rung dieser Region zu einem tieferen VerstĂ€ndnis der Pathogenese des DiGeorge-Syndroms und isolierter Fehlbildungen insgesamt beitragen kann.
Im ersten Teil der Arbeit wurden 16 Patienten mit partieller Monosomie 10p zytogenetisch und molekulargenetisch untersucht. Elf dieser Patienten zeigten einen DiGeorge-Syndrom Ă€hnlichen PhĂ€notyp, fĂŒnf Patienten wurden nicht in das DiGeorge-Syndrom-Krankheitsbild eingeordnet. Die Patienten besaĂen terminale und interstitielle Deletionen im GröĂenbereich von 13-48 cM. Mit Hilfe von FISH mit genomischen YAC-, PAC- und BAC-Sonden wurden die Bruchpunktregionen in den Patienten bestimmt. Bei einigen Patienten, bei denen DNA der Eltern vorlag, konnte auch eine Genotypisierung mit polymorphen Markern aus der Region vorgenommen werden. Mittels zweier Patienten konnte eine Haploinsuffizienzregion (DGCR2) kartiert werden, die fĂŒr den Herzfehler und den T-Zelldefekt verantwortlich sein sollte. Die Region DGCR2 ist um den Marker D10S585 lokalisiert und besitzt eine minimale Ausdehnung von etwa 300 kb. Eine genaue Genotyp-PhĂ€notyp-Analyse unter Einbeziehung von Patienten aus der Literatur zeigte jedoch, daĂ der gesamte PhĂ€notyp der partiellen Monosomie 10p nicht mit der Haploinsuffizienz nur einer Region erklĂ€rt werden konnte, sondern daĂ zumindest noch ein zweiter Locus (HDR1) deletiert sein muĂte. Dieser Locus war mit dem typischen DiGeorge-Syndrom-Symptom des Hypoparathyreoidismus assoziiert. ZusĂ€tzlich kartierten in diesen Locus noch eine sensorineurale Taubheit und Nierendefekte. Patienten mit diesen drei Symptomen leiden an einem HDR-Syndrom. Dieser zweite Haploinsuffizienzlocus HDR1 kartiert etwa 3 Mb distal zur Region DGCR2.
Im zweiten Teil der Arbeit wurde sowohl ĂŒber die Region DGCR2 als auch ĂŒber die HDR1-Region ein PAC/BAC-Contig etabliert. AusgewĂ€hlte Klone aus den Contigs wurden im Rahmen des Humangenomprojekts vom Sanger Centre sequenziert.
Der dritte Teil der Arbeit beschĂ€ftigte sich mit der molekulargenetischen Untersuchung der beiden Haploinsuffizienzregionen DGCR2 und HDR1. Es konnten 12 EST-Klone in die Region DGCR2 kartiert werden. Bei allen Klonen handelte es sich um Transkripte, die nicht zu funktionellen Proteinen translatiert wurden. Nachdem die genomische Sequenz zugĂ€nglich war, konnte eine In-silico-Analyse dieser Region durchgefĂŒhrt werden. Es handelt sich um eine sehr genarme Region. In die minimale Region DGCR2 kartiert nur das Gen NAPOR, das fĂŒr ein RNA bindendes Protein kodiert. Es wurde als Kandidatengen fĂŒr den mit dieser Region assoziierten Herzfehler und T-Zelldefekt nĂ€her charakterisiert. Eine Northern-Blot-Hybridisierung zeigte eine Expression in allen aufgetragenen Herzgeweben. Es wurden mindestens sechs verschiedene Transkripte identifiziert, was fĂŒr die Existenz mehrerer Isoformen des Gens spricht. RNA-in-situ-Hybridisierungen auf Schnitte humaner Embryos und Foeten ergaben eine Genexpression in verschiedenen Geweben beginnend von Embryos des Carnegie-Stadiums C12 bis zu 18 Wochen alten Foeten. Es wurde eine Expression im Thymus vom Carnegie-Stadium C16 an und eine Expression im Herzen bei einem Foetus der 9. Woche beobachtet. Das Expressionsmuster machte NAPOR zu einem guten Kandidatengen fĂŒr den mit der Haploinsuffizienzregion DGCR2 assoziierten Herzfehler und T-Zelldefekt. Mutationsanalysen in mehr als 100 Patienten ergaben keine Mutationen im NAPOR-Gen. Die meisten untersuchten Patienten besaĂen einen DiGeorge-Syndrom Ă€hnlichen PhĂ€notyp waren aber zytogenetisch normal. Besonderer Wert wurde auf die Anwesenheit eines Herzfehlers gelegt. Nur bei etwa 10 % der untersuchten Patienten lag auch eine Thymus-Hypoplasie vor. Ein direkter Beweis fĂŒr die Beteiligung des NAPOR-Gens am Herzfehler und/oder T-Zelldefekt bei Patienten mit partieller Monosomie 10p steht noch aus.
Die HDR1-Region konnte mit Hilfe zweier Mikrodeletionspatienten auf etwa 200 kb eingegrenzt werden. In diese Region kartiert das Gen GATA-3. Mutationsanalysen in zytogenetisch normalen HDR-Patienten zeigten in drei Patienten Mutationen, die zu einem funktionslosen GATA-3-Protein fĂŒhren. Damit wurde der Beweis erbracht, daĂ das HDR-Syndrom eine Einzelgenerkrankung ist und daĂ die Symptome Hypoparathyreoidismus,sensorineurale Taubheit und Nierendefekte bei Patienten mit partieller Monosomie 10p auf eine Haploinsuffizienz des GATA-3-Gens zurĂŒckzufĂŒhren sind.
ZusĂ€tzlich zu Patienten mit einer partiellen Monosomie 10p wurden auch zwei Patienten nĂ€her charakterisiert, die eine interstitielle Deletion auf dem Chromosom 14q11-q13 aufwiesen. Diese Region war von Interesse, da das Gen PAX-9 dorthin kartiert und homozygote Pax9 -/- Knockout-MĂ€use unter anderem eine Thymus-Hypoplasie und einen Hypoparathyreoidismus zeigen. Die MĂ€use haben zwei der drei Leitsymptome des DiGeorge-Syndroms und stellen eine Beziehung zum PhĂ€notyp der partiellen Monosomie 10p her. Die Deletionsbruchpunktregionen der beiden Patienten wurden ĂŒber eine Genotypisierung mit polymorphen Markern identifiziert. Mit Hilfe eines Dosis-Blots und einer FISH-Analyse konnte gezeigt werden, daĂ beide Patienten fĂŒr PAX-9 hemizygot waren. Beide Patienten zeigen keine Symptome des DiGeorge-Syndroms, was daraufhin weist, daĂ beim Menschen eine PAX-9-Haploinsuffizienz nicht zu einem DiGeorge-Syndrom Ă€hnlichen PhĂ€notyp fĂŒhrt.
Die in der vorliegenden Arbeit durchgefĂŒhrten Untersuchungen an Patienten mit partieller Monosomie 10p und an Patienten mit einer interstitiellen Deletion 14q11-q13 lieferten einen Beitrag zur molekulargenetischen Charakterisierung des DiGeorge-Syndroms. Der DiGeorge-Syndrom Ă€hnliche PhĂ€notyp bei vielen Patienten mit einer partiellen Monosomie 10p ist das Resultat eines Contiguous Gene Syndroms, bei dem mindestens zwei unabhĂ€ngige Regionen (DGCR2 und HDR1) auf Chromosom 10p hemizygot vorliegen mĂŒssen. Es wurde gezeigt, daĂ eine GATA-3-Haploinsuffizienz u.a. zu einem Hypoparathyreoidismus fĂŒhrt, einem der drei Leitsymptome des DiGeorge-Syndroms. FĂŒr den mit dem Syndrom assoziierten Herzfehler und T-Zelldefekt wurde mit NAPOR ein gutes Kandidatengen aus der Haploinsuffizienzregion DGCR2 identifiziert und charakterisiert. Induced pluripotent stem cell-derived neuronal cells from a sporadic Alzheimerâs disease donor as a model for investigating AD-associated gene regulatory networks
Background Alzheimerâs disease (AD) is a complex, irreversible
neurodegenerative disorder. At present there are neither reliable markers to
diagnose AD at an early stage nor therapy. To investigate underlying disease
mechanisms, induced pluripotent stem cells (iPSCs) allow the generation of
patient-derived neuronal cells in a dish. Results In this study, employing iPS
technology, we derived and characterized iPSCs from dermal fibroblasts of an
82-year-old female patient affected by sporadic AD. The AD-iPSCs were
differentiated into neuronal cells, in order to generate disease-specific
protein association networks modeling the molecular pathology on the
transcriptome level of AD, to analyse the reflection of the disease phenotype
in gene expression in AD-iPS neuronal cells, in particular in the ubiquitin-
proteasome system (UPS), and to address expression of typical AD proteins. We
detected the expression of p-tau and GSK3B, a physiological kinase of tau, in
neuronal cells derived from AD-iPSCs. Treatment of neuronal cells
differentiated from AD-iPSCs with an inhibitor of Îł-secretase resulted in the
down-regulation of p-tau. Transcriptome analysis of AD-iPS derived neuronal
cells revealed significant changes in the expression of genes associated with
AD and with the constitutive as well as the inducible subunits of the
proteasome complex. The neuronal cells expressed numerous genes associated
with sub-regions within the brain thus suggesting the usefulness of our in-
vitro model. Moreover, an AD-related protein interaction network composed of
APP and GSK3B among others could be generated using neuronal cells
differentiated from two AD-iPS cell lines. Conclusions Our study demonstrates
how an iPSC-based model system could represent (i) a tool to study the
underlying molecular basis of sporadic AD, (ii) a platform for drug screening
and toxicology studies which might unveil novel therapeutic avenues for this
debilitating neuronal disorder

Mineralogic Residence and Desorption Rates of Sorbed 90Sr in Contaminated Subsurface Sediments: Implications to Future Behavior and In-Ground Stability
The project is investigating the adsorption/desorption process of 90Sr in coarse-textured pristine and contaminated Hanford sediment with the goal to define a generalized reaction-based model for use in reactive transport calculations. While it is known that sorbed 90Sr exists in an ion exchangeable state, the mass action relationships that control the solid-liquid distribution and the mineral phases responsible for adsorption have not been defined. Many coarse-textured Hanford sediment display significant sorptivity for 90Sr, but contain few if any fines that may harbor phyllosilicates with permanent negative charge and associated cation exchange capacity. Moreover, it is not known whether the adsorption-desorption process exhibits time dependence within context of transport, and if so, the causes for kinetic behavior
